Question: If another region is added, meaning there is more than one, are the fees for the second or third region charged again?

No, there's no additional fee for deploying to extra regions. You pay for the combined resource usage of all replicas - each replica is billed independently for the CPU, RAM, and egress it uses, at the same rates regardless of region.

Sorry, I didn't understand you or I didn't phrase the question correctly. If I currently have an application deployed in US East (Virginia, USA) and I add a second region, for example, US West (California, USA), will I be charged for both regions?

Yes, you pay for the resources each replica uses - if you run replicas in both US East and US West, you're billed for the CPU, RAM, and egress consumed by each, at the same rates with no regional surcharge.
